What this library explains

These are mechanisms investigated in ingredient research and discussed in the ENDOLLS manuscript. They explain the formulation rationale; they are not a claim that Advanced Duo produces every effect described. Each explanation identifies the study setting and its limits.

Cell stress and signaling

Rhodiola rosea

3% salidroside · 3% rosavins

Salidroside is one of the compounds the manuscript follows most closely. Rosavins are another marker group identified on the current label.

The mechanism being investigated

In experimental systems, salidroside has influenced HIF-1α, a response to low oxygen, and LOXL2, an enzyme involved in tissue remodeling. Other experiments connect it with Nrf2 antioxidant signaling and NF-κB inflammatory signaling.

What was actually studied?

Chen and colleagues studied pancreatic cancer cells and a mouse tumor model. Tang and colleagues studied chemically induced lung injury in rats. These help explain the manuscript’s interest in the pathways.

How this relates to Advanced Duo

Neither study tested Advanced Duo or demonstrated an endometriosis benefit in people. Cancer and lung models cannot be treated as interchangeable with endometriosis.

Smooth-muscle activity

Valerian root

0.8% valerenic acid

Valerian contains several constituents. The label standardizes valerenic acid; whole extracts and individual molecules are not equivalent.

The mechanism being investigated

The manuscript explores how valerian constituents may affect the signals controlling muscle contraction. Laboratory work on isolated human uterine tissue found that valerian extracts reduced contraction as concentration increased.

What was actually studied?

A separate randomized trial studied 100 students with primary menstrual pain using a different dose and schedule. It was not an Advanced Duo trial. The manuscript also cites Valeriana wallichii animal-tissue work—a species-specific result that cannot establish the action of this label’s extract.

How this relates to Advanced Duo

These findings do not show that the Duo prevents reflux, implantation, or disease progression. Tissue-bath concentrations cannot be converted directly to a capsule dose.

Inflammatory messengers

Salicin is a plant compound found in willow bark. It is chemically distinct from aspirin.

The mechanism being investigated

The manuscript’s interest includes NF-κB, a regulator of inflammatory gene activity. In retinal endothelial cells, researchers tested salicin against an IL-1β inflammatory challenge and observed reduced NF-κB activation and several inflammatory signals.

What was actually studied?

This provides a specific laboratory example of the pathway discussed in the formulation rationale.

How this relates to Advanced Duo

It does not prove that the oral extract interrupts an estrogen feedback loop, prevents lesions, or produces the same effects in people. Adding beta-glucan does not establish protection from adverse effects.

Estrogen-receptor signaling

DIM is associated with compounds in cruciferous vegetables. Its biological effects depend on concentration and cell type.

The mechanism being investigated

Vivar and colleagues observed ERβ-linked gene activation involving SRC-2, a protein that helps regulate gene transcription. DIM did not need to bind directly to ERβ in that experiment. This is more specific than simply calling it an estrogen blocker.

What was actually studied?

Another cell study found ERα activation at low concentrations. The textbook also describes altered receptor patterns in endometriosis, making context especially important.

How this relates to Advanced Duo

The cited work does not establish that Renewal normalizes hormones or safely replaces prescribed hormonal treatment. Do not change medication on the basis of these mechanisms.

Inflammation and tissue remodeling

Rose hip contains multiple compound groups. A powder, a purified galactolipid, and a polyphenol-standardized extract are different preparations.

The mechanism being investigated

The manuscript discusses cytokines and MMPs—enzymes involved in remodeling tissue. Schwager and colleagues tested rose hip powder and a galactolipid in immune and cartilage cells and observed changes in inflammatory mediators and MMP expression.

What was actually studied?

This offers a laboratory basis for investigating these pathways, with the tested preparation clearly identified.

How this relates to Advanced Duo

It does not show that the labeled extract prevents lesions, repairs nerves, thickens myelin, or treats endometriosis. Results from cartilage models are not clinical results for this regimen.

Fibrin breakdown

Nattokinase

Nattokinase is an enzyme studied for fibrinolytic activity. FU describes activity; milligrams describe weight.

The mechanism being investigated

Fibrinolysis is the breakdown of fibrin, a protein involved in blood clots. In a small human crossover study, a single 2,000 FU dose changed several markers related to clotting and fibrin breakdown. Changes remained within normal ranges.

What was actually studied?

That study involved 12 healthy young men and measured blood markers over eight hours. It provides human evidence about a specific biological process.

How this relates to Advanced Duo

Fibrin is not interchangeable with collagen, fibronectin, or a mature pelvic adhesion. The study did not demonstrate that nattokinase dissolves lesions or scar tissue.

The digestive environment

Oat beta-glucan

Oat beta-glucan is a soluble polysaccharide. Its linkage pattern, molecular size, and preparation matter; yeast beta-glucan is not the same ingredient.

The mechanism being investigated

Oat beta-glucan can form viscous solutions, which is part of the manuscript’s digestive rationale. A 30-day study in 48 people with chronic gastritis compared high- and low-molar-mass preparations with placebo.

What was actually studied?

The high-molar-mass preparation showed favorable changes in gastric mucosal damage and selected biochemical measures.

How this relates to Advanced Duo

This was not a trial in endometriosis and did not test the Duo’s dose or formula. It does not show that 90 mg protects against NSAID-related injury or makes other ingredients risk-free.
